There are over fifty modified bases in tRNAs. Are the nucleotides modified before or after incorporation into the tRNA?

What will be an ideal response?

Ans: They are modified after transcription of the tRNA. If the modifications occurred on free nucleotides then all cellular RNAs would contain them. Modifying enzymes alter the nucleotides at specific positions in tRNAs by recognizing both the sequence and the structure of those regions.
