Yamamoto cloned the mHTT gene behind an inducible promoter that responded to doxycycline. When the mice were fed water containing doxycycline, the mHTT gene was turned on and disease progression commenced. When doxycycline was removed from the water, the mHTT gene was turned off and disease progression halted. Yamamoto monitored the aggregates of mHTT and determined that if the gene expression was arrested early enough, the disease state could be reversed by normal cellular functions.
