Answer: Both the MPN and MF procedures provide information on the concentration of coliforms and E. coli. in the procedure for determining the MPN, a water sample is added into a defined liquid culture medium where it is incubated to observe growth (and therefore presence) of particular organisms. The concentration is often indirectly determined using the number of liquid cultures positive for a particular organism along with an MPN table. The MF procedure involves filtering a water sample such that the microbial cells are trapped onto the filter, and the filter is placed onto a differential and selective growth medium such as a eosin-methylene blue (EMB) agar plate. The agar plate is incubated and colony-forming units are directly counted.
