The ob mouse is a genetic knockout that does not have the OB gene. The OB gene is responsible for making leptin, a long-term satiety signal. Given that these animals do not have this long-term satiety signal, they have a tendency to overeat, have low metabolism, and do not engage in exercise. Over time, they become obese. Interestingly, injection with leptin will return these mice to their normal weight over time. This demonstrates that leptin is an important signal for weight maintenance.
