Describe the Southern blotting procedure for analyzing DNA

A piece of DNA to be analyzed is digested by restriction enzymes. The resulting fragments are separated by
agarose electrophoresis. The gel is soaked in an alkaline solution to convert the DNA to single-stranded form. Then
a nitrocellulose membrane is laid over the agarose. The DNA is transferred to the nitrocellulose by "blotting." The
blotting traditionally was done by capillary action, but newer techniques employ either a vacuum or an electrical
field to speed the process. Since the DNA is now single-stranded and fixed to the nitrocellulose, specific sequences
can easily be detected using nucleic acid probes.
