What do the "puffs" of polytene chromosomes represent?

Ans: A puff is a region of the polytene chromosome where the chromosome fibers have become unraveled from their tight packing. This is correlated with gene expression in that region. The structure is relaxed in this region to allow access of proteins necessary for transcription.
