Describe the principle of TaqMan real-time PCR

In TaqMan real-time PCR, the PCR reaction proceeds as usual except a labeled probe is contained in the reaction
mixture. The probe anneals to a sequence within the PCR product, so unless PCR product is formed, it has nothing
to which to bind. The probe is labeled at 5' end with a fluorescent molecule and at the 3' end with a quencher
molecule. One of the properties of DNA polymerase is 5' →3' exonuclease activity when a piece of DNA is being
synthesized. If the probe has bound to the target DNA in the PCR product, during the next extension cycle, the
DNA polymerase will cleave off the nucleotides in the probe. This separates the fluorescent molecule from the
quencher molecule, and fluorescence is detected. Thus, increased levels of fluorescence correspond to increased
amounts of PCR product.
