The Cowan 1 strain of Staphylococcus aureus contains a large amount of protein A on the cell surface. Protein A binds
to the Fc portion of IgG, leaving the antigen binding portion available. Thus, antibodies can be attached to the
Staphylococcus aureus cells, and when the corresponding antigen is present, visible agglutination will occur. This
causes a problem, however, in that any IgG in the specimen is capable of also binding to the Staphylococcus aureus
cells. Specimens containing rheumatoid factor (an IgM antibody against IgG) will visibly agglutinate the
Staphylococcus aureus cells, causing a false positive.
