Ans: The gel matrix itself can determine resolution. Use of agarose allows resolution of fragment size differences of about 50 bp, while the use of a polymer matrix (such as polyacrylamide) allows separation of fragments that differ in size by only 1 bp. Resolution is also higher by using thinner gels; a very fine capillary tube provides high-resolution fragment separation.
