Answer: To enrich for heterotrophic sulfate-reducers, the media must contain an organic source of carbon, a source of N, P, and trace elements, and sulfate. The growth conditions must also be anaerobic, as most sulfate-reducers are obligate anaerobes and marine sediments are mostly depleted or devoid of oxygen. The carbon source should be hydrocarbons similar to those found in crude oil or very small amounts of crude oil. Lastly, the bottle or tubes should be incubated in the dark to prevent the growth of anoxygenic phototrophs.
