The origin of a plasmid that contains a selectable marker such as AmpR was removed. The plasmid was ligated to fragments of DNA from the bacteria of interest and the resulting recombinants were transformed into the bacteria. If the transformed bacteria grew on ampicillin media then they contain a functional origin from the bacteria of interest.
