An adequate number of pluripotent stem cells must be infused into a recipient to restore normal hematopoietic
capacity. Older techniques measured mononuclear cells in the expectation that a small percentage of mononuclear
cells would be pluripotent stem cells, although there was no guarantee that this was so. CD34 is one of the earliest
markers expressed in hematopoietic progenitors, and since it is lost at later stages of maturity, it is currently
considered the best marker for the ability of a harvested cell population to repopulate a bone marrow. All human
white blood cells express the CD45 marker, so a count is done of cells bearing both the CD45 and CD34 markers.
This count can then be used to estimate how much must be infused to transfer an adequate number of progenitor
cells.
