Boon's group established a stable melanoma cell line from a patient whose tumor cells expressed several tumor
associated antigens, including antigen E. The group isolated CTLs from the patient that were reactive against
antigen E and that could lyse the melanoma cells grown in culture. When the CTLs were cocultured with the
melanoma line, a variant cell line emerged that lacked antigen E. A DNA cosmid library was constructed from the
original cell line that possessed E, and fragments of DNA were inserted into the E negative cell line until CTL
recognition was restored. The DNA fragments that restored E antigen were subsequently honed down to the
MAGE-1 gene that encoded for a specific 9 amino acid epitope designated as "E." In this way, a precise tumor
associated antigen and its encoding gene were definitively identified.
