Answer: The primary concern for codon usage is that one organism might natively make a large quantity of a particular amino acid, so proteins rich in this particular amino acid can maintain a high level of translation, yet the cloning host does not make high levels of the amino acid. This results in unexpectedly low levels of the engineered gene product.
