Explain how flow cytometry and hematology cell counting are both used to obtain

absolute CD4+ lymphocyte counts.

What will be an ideal response

Flow cytometry results are typically reported in percentages. For example, if 1000 cells are analyzed and 100 of
them have the CD4 marker, then the value reported is 10%. To convert this value to a count per volume of blood,
cell counters must determine absolute lymphocyte counts per volume of blood. For example, if there are 5000
lymphocytes/uL of blood, and 20% of all lymphocytes possess the CD4 marker, then the CD4 count is 5000 x 20%
or 1000 CD4 cells/uL.
