Answer: Answers will vary, but a common method is to digest the DNA sample and clone the fragments into a vector. The particular vector will vary depending on the size range of the digested fragments such as plasmids, cosmids, BACs, HACs, YACs (in ascending order of insert size capacity). If the library is to be use for functional analysis, a vector also must contain a promoter neighboring the upstream insertion site.
