Ans: In prokaryotes, the assembly of the 30S subunit occurs directly over the AUG via base pairing between the ribosome-binding site and the 16S rRNA. There is no complementarity between the sequences surrounding the AUG and the 18S rRNA of eukaryotes. Instead, initiation factors bind both the 5? methyl G cap and the poly(A) tail, and assemble with the 40S subunit at the 5? methyl G cap. The 40S subunit scans the mRNA from the 5? end until ribosomal proteins recognize the initiator region, or the ribosome-binding site. This sequence is NNNPuNNAUGG, where AUG is the initiator codon.
