First, efforts are made to standardize the amount of starting RNA material in each sample. Since this can be difficult
due to differences in samples and imprecision in extraction procedures, mRNA from a housekeeping gene is
usually also quantified simultaneously. A housekeeping gene is defined as a gene that is consistently expressed
over time by all cells such that certain levels can be expected and can be used as a measure of how well the
procedure has performed. The beta-actin gene transcript is often used in eukaryotes while the 16S rRNA is often
used in prokaryotes.
