How did Clark and Margulies screen for recombination deficient mutants?
What will be an ideal response?
They spread Hfr cells that were leu- on media lacking both leucine and adenine. They then transferred their mutagenized ade- F- cells onto the lawn of Hfr cells. The Hfr cells mated with the F- cells and many of them generated leu+ade+ recombinants that grew on the selective media. The mutants that did not grow lacked recombination capability.
