Since aconitase is involved in iron utilization, its expression should be low when iron levels are low. When iron levels are high, aconitase levels should be high as well. Since IRPs have low affinity for IRE regulatory sequences when bound to iron, the translation of aconitase mRNA should be inhibited when IRPs are not bound by iron. Therefore, the IRE should be in the 5' UTR so that translation is blocked when the IRP is bound.
