Answer: Specimen preparation in tissue processing is a technique by which the tissue from a source are processed so that they can be sliced effectively in a microtome for various purposes.

The type of specimen preparation which involves dehydrating the tissue is the paraffin block method. Here, the tissue is completely fixed to a histological wax and specimen is created.

The steps involved are :

Collection of the specimen, fixing of specimen , dehydration , infiltration of the wax , blocking out.

Dehydrating these tissue is carried out to remove the water molecules so that they can be effectively infiltrated into the paraffin wax. If not dehydrated, paraffin being hydrophobic will not immerse the specimen as per requirement. Dehydration process is often carried out in the presence of alcohol where the fixed specimen is allowed to stand in an ethanolic solution until the solution becomes free of water.
